4-Azidosalicylic acid NHS ester is an amine-reactive heterobifunctional cross-linker. At neutral or slightly basic pH (7-9), it rapidly attacks solvent accessible lysine-NH2 groups, N-termini and amines and installs the arylazido group that can both undergo a radioiodination and subsequently be transformed to reactive nitrene by irradiation with short wavelenghts (250 nm). The in-situ generated nitrene reactive intermediate is highly reactive and immediately gets captured by surrounding nucleophiles.
